Do you need a power supply?

No. It's a passive footswitch, no power required.
Is it only compatible with Ampero?

No. It works with any pedal, multi-effects device, or amp that has an external footswitch input.
What is the difference with a classic footswitch?

The Switch+ is momentary: it sends a short pulse, perfect for digital commands (presets, tap tempo, etc.).
Can I freely assign both switches?

Yes. In your Ampero multi-effects settings, you can assign a function to each switch.
